Bij de Hollandse Maatschappij ter Fabricatie van Gecondenseerde melk was hij in dienst als scheikundige. Hij was van plan om over enige tijd, evenals zijn broer Henk, naar Nederlands Indië te vertrekken. Op een avond zou hij vanuit de fabriek nog gauw even een brief posten. Hij liep vlug tussen een paar stilstaande goederenwagons van een stoomtram door. Juist op het moment dat hij tussen de buffers was, werd tegen één van de wagons, door enkele arbeiders, een andere wagon geduwd. Hij werd zwaar gewond in het gemeenteziekenhuis aan de Coolsingel 63 in Rotterdam opgenomen. Enkele uren later is hij aan zijn zware verwondingen overleden. Op 21 mei 1917 is hij onder grote belangstelling begraven in Axel. Ds. van Dis uit Zaamslag leidde de begrafenis. Hij had de overtuiging dat "Gods woordte allen tijde troost schenkt en zelfs de grootste droefheid lenigt". De moeder van Co was zo overstuur dat ze niet bij de begrafenis aanwezig kon zijn. Zijn vader nam afscheid met de woorden: "Tot weerziens mijn zoon". Na zijn dood hing er een groot portret van hem boven de deur in de pastorie in Axel. Zijn graf was tijdens mijn bezoek aan Axel in 1985 nog aanwezig, maar was in zeer slechte staat. Op de avond van het ongeluk schrok zijn moeder wakker en zei tegen haar man: "Co is dood!". Haar man kalmeerde haar en ze gingen weer slapen. Enige tijd later kregen ze een telegram met de boodschap: "Zoon ernstig ziek, komt direct". Vele tientallen jaren na zijn dood (halverwege jaren '70), werd mijn grootmoeder Hugenholtz-Lehmkuhl benaderd door een vrouw die informeerde naar Co Hugenholtz. Zevertelde dat Co haar vriend was geweest. Ze gingen vaak grote stukken samen wandelen. Hij was echter plotseling niet meer op komen dagen, en sindsdien had ze niets meer van hem gehoord. Pas toen hoorde deze vrouw dat hij niet meer was gekomen omdat hij verongelukt was.
