Johan de Mepsche woonde op de borg Piloersma te Den Ham. Hij was hoofdeling te Groningen, evenals zijn gelijknamige neef Johan (IV.2). In tegenstelling tot zijn neef was hij staatsgezind. Door de Spaanse overheersing in de stad Groningen was hij genoodzaakt om te vluchten, hij overleed in ballingschap.
Zijn schoonfamilie Jensema leefde al enige eeuwen op de Jensemaborg te Oldehove. Zijn schoonvader Rembt Jensema overleed eveneens in ballingschap, na 1580.

They got married in the year 1567, he was 37 years old.
