Martinus behaalde in Leuven zijn magistergraad in de wijsbegeerte, waarna hij theologie ging studeren. Hij gaf onderwijl les in Latijn en Wijsbegeerte. In het jaar 1515 werd hij tot priester gewijd. Martinus was het niet eens met de humanist Erasmus, die kritiek uitte tegen het gebruik van Latijnse Bijbelteksten. Deze kritiek vond plaats middels vele briefwisselingen. Uiteindelijk legden Dorpius en Erasmus hun geschil weer bij. Dorpius kwam echter wel in moeilijkheden met zijn faculteit, toen hij een humanistische benadering van de theologie verdedigde. Toch werd hij in 1517 tot decaan gekozen van het H. Geestcollege. Vanaf 1519 zijn er geen publicaties meer van zijn hand verschenen, waarschijnlijk om verdere conflicten te vermijden. In 1523 was hij rector van de universiteit. Op 31 mei 15 25 stierf Martinus Dorpius te Leuven. Zijn graf in het Leuvense karthuizerklooster werd gesierd door een grafdicht van Erasmus.
