    Gillis ging in 1623 naar Engeland; medewerker van Cornelis Jacobsz. Drebbel; reisde met zijn broer Abraham in 1624 en 1625 in Frankrijk en Italie; richtte in 1635 met zijn broers Dr. Johannes Sibertus en Abraham onder de naam Gebroeders Kuffeler een compagnieschap in het scharlakenverven op, welke te Stratford Bow aan de Lea een ververij had; hij was in 1636 conducteur van de vuurwerkers op de vloer van admiraal Philips van Dorp; In 1639 begaf hij zich naar Amsterdam waar hij zijn intrek nam bij zijn oom Abraham, die aldaar in het huis In de Groen Vijffhoeck aan de Keizersgracht woonde; in 1641 woonde hij aan de Rozengracht; op 14 april 1641 kocht hij de hofstede Rustenburgh gelegen buiten de Reguliersgracht aan de oostzijde van de Amstel welke hij op 4 juli 1650 weer verkocht. Hij leidde de scharlakenververij van de Gebroeders Kuffeler op het landgoed Hulkestein onder Arnhem; op 1 sept. 1648 verkreeg hij van de Magistraat van Arnhem het recht het stadswapen aan de op Hulkestein geverfde lakens te hechten. Op 23 april 1649 leende hij aan de stad Oppenheim 1200 rijksdaalders, welke schuld nimmer is gedelgd ondanks daartoe aangewende pogingen van Mr. Karel Rudolpn van Kuffeler, Philip Hack en Jacob Jan de Blocq van Kuffeler; in 1658 was hij de hoofdparticipant van de West Indische Compagnie en woonde toen aan de Prinsengracht tegenover de Noorderkerk.
