Op zijn 19e trouwde hij met de behangersdochter Aaltje Noort. Jesajas had vele beroepen (venter, handelsreiziger, toneelknecht, muzikant, artiest). Zijn gezin, dat uiteindelijk uit vijf zonen en een dochter bestond (drie dochters stierven kort na de geboorte), voerde hij vanuit zijn geboorteplaats Groningen over Den Bosch en Rotterdam in 1901 naar Amsterdam. Volgens archiefstukken zou het gezin ook nog enkele keren kort in Antwerpen hebben gewoond. In Amsterdam was Jesaja Ruben naast hyperactief ook vaak bedlegerig. De armenzorg en de Joods Israëlische gemeente moesten dan bijspringen. In 1910 overleed Jesajas Ruben vrij plotseling op 23 september.
