    Die Ersterwähnung des Ortes erfolgte am 25. Mai 1468.[1] Der Ort entstand um das ursprünglich zur Grafschaft Stolberg gehörige amtssässige Allodialrittergut, das bis 1815 im Amt Sangerhausen des Königreichs Sachsen lag und dann an den Kreis Sangerhausen der preußischen Provinz Sachsen gelangte. Mit diesem Amt war Agnesdorf von 1657 bis 1748 dem Fürstentum Sachsen-Weißenfels unterstellt.


    Am 9. August 1798 erwarb das bis dahin amtssässige Rittergut die Schriftsässigkeit. Dessen Besitzer waren: Grafen zu Stolberg, v. Hopfgarten (bis 1572), v. Hacke (ab 1572), v. Wackernhagen (bis 1748), v. Worbis (ab 1748/52), von Linsingen (bis 1784), v. Bülzingslöwen, Schlitt (beide ab 1787), v. Westernhagen, Schönstedt, v. Kropf. Weiterhin besaß die adlige Familie Knauth Knutonen auf Questenberg von 1463 bis 1670 Güter im Ort. Das Rittergut war bis 1848 gleichzeitig Sitz eines Patrimonialgerichts.


    In der Familie v. Hacke war noch der ursprüngliche Name Angsdorf üblich.
