Overeenkomstig het testament van zijn vader gingen Neumark en andere gebieden als markgraafschap Brandenburg-Küstrin naar zijn broer Johan. Onder zijn bewind werd in 1539 de reformatie ingevoerd in Brandenburg. Joachim koos daarmee de zijde van de broers van zijn moeder en zijn oom Albrecht van Brandenburg-Pruisen. In 1542 liet hij de bouw van het jachtslot Grunewald aanvatten. Joachim hield een omvangrijke hofhouding en liet bij zijn overlijden grote schulden na.
