Hij volgde zijn vader in 1753 op en sloot in 1783 de Erbverein met Nassau-Saarbrücken, Nassau-Usingen en Nassau-Dietz (Oranje-Nassau) die de eenheid en onvervreemdbaarheid van Nassau en het eerstgeboorterecht erkende en bepaalde dat de verschillende linies bij uitsterven van een ervan elkaars ambten zouden erven (zoals na de dood van koning Willem III in Luxemburg zou gebeuren).

Op 5 maart 1760 huwde hij te Den Haag Carolina van Oranje-Nassau (1743-1787), zuster van Willem V. Aangezien zijn echtgenote in het geval van een minderjarig overlijden van Willem V in aanmerking kwam voor het stadhouderschap, vestigden zij zich in Den Haag aan het Korte Voorhout in een door Pieter de Swart ontworpen stadspaleis, de huidige Koninklijke Schouwburg. Nadat Willem V meerderjarig was geworden, resideerden Karel Christiaan en Carolina te Weilburg en Kirchheimbolanden.

Hij werd generaal der infanterie in Staatse dienst, gouverneur van Bergen op Zoom en gouverneur van Maastricht (1773-1784). In 1787 voerde hij vergeefs onderhandelingen met de patriotten.

In het jaar van zijn dood sloot hij een morganatisch huwelijk met Barbara Giessen von Kirchheim. Hij stierf in 1788 en werd opgevolgd door zijn oudste nog levende zoon Frederik Willem.
