    Hij is trekschuitschipper (1817), molenaar (1822), grutter (1830), landbouwer, koopman (1840) en predikant (1844). Waarschijnlijk heeft hij als trekschuitschipper in Dokkum zijn eerste vrouw Jantje Hindriks Boersma leren kennen. Hij gaat in Dokkum wonen, maar in 1822 vestigt hij zich in Stroobos als boekweitmolenaar. Op 2 juni 1822 is hij met attestatie overgekomen uit Dokkum. Daar komt hij in aanraking met de Afscheidingsbeweging, waaraan hij actief gaat deelnemen. Ds. Hendrik de Cock wordt bij zijn bezoeken aan Stroobos altijd hartelijk ontvangen in het huis van de familie Drukker. Op de eerste lidmatenlijst van de Afgescheiden Gemeente van Grootegast van 20 juni 1835 staan ook de namen Derk Derks Drukker en zijn vrouw Antje Seits Denkema. In 1840 verhuist Derk Derks Drukker naar Ulrum, waar hij al spoedig ouderling wordt. Hij gaat ook voor in kerkdiensten in Ulrum en enkele Friese plaatsen. Na twee mislukte pogingen om dominee te worden, wordt hij toegelaten tot het predikambt. Op 11-9-1844 wordt hij bevestigd tot predikant in zijn vroegere woonplaats Dokkum. In 1848 vertrekt hij naar zijn nieuwe standplaats Koudum-Hindeloopen, waar hij in 1852 komt te overlijden.
