Over het middeleeuwse Popkensburg is nauwelijks iets bekend. Men veronderstelt dat het slot gesticht is door ene Jan van Popkensborch. Wat wel vast staat, is dat slot en heerlijkheid in de veertiende eeuw in het bezit zijn van de heren van Borssele. De eerste eigenaar uit dit geslacht, Claes van Borssele, liet het kasteel in 1353 verbouwen. In 1426 werd Adriaan van Borssele de nieuwe eigenaar van Popkensburg. Deze bezat ook het kasteel te West-Souburg waar hij aan het einde van zijn leven woonde. Vermoedelijk is Popkensburg sindsdien door een slotbewaarder bewoond. Door vererving kwam het vervolgens in handen van de heren van Veere. Maximiliaan van Bourgondië (1514-1558), die in 1553 nog een verbouwing op Popkensburg liet uitvoeren, overleed berooid in 1558, waarna heerlijkheid en kasteel publiek verkocht werden. Philibert van Serooskerke en Arend van Valckensteyn werden ieder voor de helft de nieuwe eigenaar. Philibert en Arend waren Spaansgezind, zodat Popkensburg tijdens het beleg van Middelburg in de jaren 1572-1574 een sterke Spaanse bezetting had en het kasteel niet door de Geuzen werd verwoest. De stad Middelburg verwierf de heerlijkheid Popkensburg in 1612. Het kasteel werd in 1631 door Walter Fourmenois gekocht. Bij zijn overlijden in 1653 ging het kasteel over op zijn zuster Catharina, echtgenote van Pieter Boudaen Courten. In 1679 werd ook de heerlijkheid Popkensburg door Johan Boudaen Courten, inmiddels heer van Popkensburg, aangekocht zodat kasteel en heerlijkheid weer in één hand waren. Inmiddels werd de heerlijkheid aangeduid als Sint-Laurens. Kasteel en heerlijkheid kwamen in 1762 in bezit van Anna Sara Boudaen Courten, gehuwd met Jacob van Citters.
