In 1918 behaalde hij het diploma https://nl.wikipedia.org/wiki/Civiel_ingenieur">civiel ingenieur. Vanaf 1918 was hij als assistent in de https://nl.wikipedia.org/wiki/Geodesie">geodesie verbonden aan de Technische Hogeschool. In 1926 volgde zijn benoeming tot https://nl.wikipedia.org/wiki/Hoogleraar">hoogleraar in het https://nl.wikipedia.org/wiki/Landmeetkunde">landmeten, waterpassen en https://nl.wikipedia.org/wiki/Geodesie">geodesie. Zijn speciale interesse lag op het vlak van de https://nl.wikipedia.org/wiki/Fotogrammetrie">fotogrammetrie. In 1948 organiseerde Schermerhorn het negende congres van de https://nl.wikipedia.org/w/index.php?title=ISPRS&action=edit&redlink=1">ISPRS (International Society for Photogrammetry and Remote Sensing) in Den Haag, waar werd besloten tot het opzetten van het International Training Centre for Aerial Survey (ITC), een opleidingsinstituut met een focus op ontwikkelingslanden, thans https://nl.wikipedia.org/wiki/International_Institute_for_Geo-Information_Science_and_Earth_Observation">International Institute for Geo-Information Science and Earth Observation geheten. In 1950 besloot de Nederlandse overheid het ITC daadwerkelijk op te richten. Schermerhorn werd de eerste rector van het ITC in Delft. Van 1951 tot 1965 was hij directeur van het Internationaal Instituut voor Luchtkartering te Delft. Schermerhorn nam resoluut stelling tegen de opkomende https://nl.wikipedia.org/wiki/Nationaal-Socialistische_Beweging">NSB. Schermerhorn was in juni 1935 een der oprichters van de buitenparlementaire beweging https://nl.wikipedia.org/wiki/Eenheid_door_Democratie">Eenheid door Democratie. In 1938 werd hij na een bestuurscrisis voorzitter op verzoek van https://nl.wikipedia.org/wiki/Jan_Goudriaan">Jan Goudriaan. Ook was hij jarenlang bestuurslid van de https://nl.wikipedia.org/wiki/VPRO">VPRO. Tijdens de Tweede Wereldoorlog, van 1942 tot 1943 werd Schermerhorn geïnterneerd in het https://nl.wikipedia.org/wiki/Kamp_Sint-Michielsgestel">kamp Sint-Michielsgestel. Hier was hij een van de zogenaamde 'https://nl.wikipedia.org/wiki/Heeren_Zeventien_(politiek)">Heeren Zeventien'. Een groep van tien gegijzelden in Sint-Michielsgestel besloot tot de oprichting van de https://nl.wikipedia.org/wiki/Nederlandse_Volksbeweging">Nederlandse Volksbeweging zodra de bezetting voorbij was. Na zijn vrijlating in december 1943 dook hij onder. In https://nl.wikipedia.org/wiki/1944">1944 ontsloeg de Duitse bezettende macht hem als hoogleraar. In de bezettingstijd was hij een verbindende figuur in de https://nl.wikipedia.org/wiki/Nederlands_verzet_in_de_Tweede_Wereldoorlog">Nederlandse illegaliteit. Hij speelde een centrale rol in tal van overlegorganen, die de opbouw van de Nederlandse maatschappij na de bevrijding wilden vormgeven. Na de oorlog nam hij kort zijn hoogleraarsstoel weer in aan de Technische Hogeschool Delft.

Van 24 juni 1945 tot 3 juli 1946 was Schermerhorn minister-president en minister van Algemene Oorlogsvoering van het Koninkrijk in het https://nl.wikipedia.org/wiki/Kabinet-Schermerhorn-Drees">Kabinet-Schermerhorn-Drees. Al snel nadat de wederopbouw was begonnen werd Schermerhorns kabinet verrast door het uitroepen van de https://nl.wikipedia.org/wiki/Republik_Indonesia">Republik Indonesia. Nadat zijn kabinet viel hielden de katholieke leider https://nl.wikipedia.org/wiki/Louis_Beel">Beel en partijgenoot Drees Schermerhorn buiten de formatie en het nieuwe kabinet. Vervolgens werd hij benoemd tot voorzitter van de https://nl.wikipedia.org/wiki/Commissie-Generaal_voor_Nederlands-Indi%C3%AB">Commissie-Generaal voor Nederlands-Indië. Deze functie oefende hij uit in 1946 en 1947. De door Schermerhorn nagestreefde partijpolitieke vernieuwing verliep teleurstellend. De verzuilde structuren bleken hechter dan verwacht.
