    De stamreeks begint met Maes Molengrave die tussen 1416 en 1433 schepen van Loon op Zand was. Een nazaat, Willem Jansz van Loon, werd bewindhebber van de VOC-kamer te Rotterdam.

    De familie vluchtte waarschijnlijk in december 1577 in paniek uit 's-Hertogenbosch, nadat duidelijk was geworden dat het stadsbestuur niet overging tot de erkenning van de Unie van Utrecht en Alexander Farnese, de hertog van Parma om bescherming vroeg. De calvinisten in ’s Hertogenbosch wilden aansluiting bij de Unie van Utrecht, maar het katholieke stadsbestuur verzoende zich met Parma, waarna 1/5 van de bevolking de stad verliet. Een aantal Van Loon's kwam via Rotterdam in Amsterdam terecht.
