      Haar grootvader aan vaderskant was George van Brunswijk-Calenberg en haar grootouders aan moederskant was koning Frederik V van de Palts en koningin Elizabeth Stuart, die een dochter was van koning Jacobus I van Engeland. Sophie Charlotte had meerdere broers en zussen waaronder Georg Ludwig die na de dood van koningin Anna van Groot-Brittannië in 1714 koning van Groot-Brittannië werd als George I. Ze was een tante van koning George II van Groot-Brittannië en van koningin Sophia Dorothea van Hannover die later zou trouwen met haar zoon de latere koning Frederik Willem I van Pruisen.

      Er werd gezegd dat Sophie Charlotte zou trouwen met koning Lodewijk XIV van Frankrijk na de dood van diens vrouw koningin Maria-Theresia van Spanje maar dit was enkel een roddel. In 1684 trad Sophie Charlotte in het huwelijk met Frederik van Pruisen de zoon van hertog Frederik Willem van Brandenburg en de latere koning in Pruisen.

      Sophie Charlotte is vooral bekend gebleven door haar vriendschap en haar briefwisseling met Gottfried Leibniz.
