He became a retainer of the local magnates, the In 1469 he was present at the Siege of Caister Castle, and in 1471 as a member of the victorious Yorkist forces he was knighted by King Edward IV on the field at the Battle of Tewkesbury. Rising to be a senior member of the council of John de Mowbray, 4th Duke of Norfolk by 1476, as a member of the Royal Household, in 1479 He was appointed Knight Marshal of the Marshalsea Court,an office for life which passed to his son Thomas in 1491.The Marshalsea Court (or Court of the Marshalsea, also known as the Court of the Verge or the Court of the Marshal and Steward) was a court associated with the Royal Household in England. Associated with, but distinct from, the Marshalsea Court was the Palace Court.

In July 1483 he was present at the coronation of King Richard III, but despite marks of royal favour his loyalty became suspect when two of his sons, William and Thomas, joined the rebellion of Henry Stafford, 2nd Duke of Buckingham in October. Some of his lands were seized by Thomas Hopton on the King's orders, but he secured a free pardon in March 1484. By the end of that year, he was out of favour again and sought sanctuary in the City of Gloucester, where he remained until after Richard's defeat and death at the Battle of Bosworth in August 1485.
Later that year he petitioned Parliament for, and regained, his office of Knight Marshal.

The earliest recorded member of the Brandon family was one William Brandon (circa 1425 - 1491) of Wangford, Suffolk, and Soham, Cambridgeshire, who, during the reign of King Edward IV, became Knight Marshal of the Marshalsea prison. Brandon married Elizabeth Wingfield (d. 28 April 1497), daughter of Sir Robert Wingfield and Elizabeth Goushill, the couple had three sons and seven daughters.

Charles Brandon Duke of SuffolkWilliam's eldest son, Sir William Brandon (1456 - 22 August 1485) of Soham, married Elizabeth Bruyn, the daughter and co-heiress of Sir Henry Bruyn of Essex, and widow of Thomas Tyrrell of Heron. William and Elizabeth had two sons and a daughter, William Brandon who died before 1500, Charles Brandon, later 1st Duke of Suffolk (circa 1484 - 24 August 1545) and Anne Brandon, who married firstly Sir John Shilston, and secondly Sir Gawain Carew.
