Hij huwde 1e Haastrecht 30 januari 1771 (Jacob betaalt 15 gulden aan de gaarder van Haastrecht en Neeltje 15 gulden aan de gaarder van het Land van Steijn) met Neeltje van Grieken, geb. Stein (bij Haastrecht), ged. Haastrecht 1 maart 1744, begr. Stein 24 november 1771 (aangeg. impost Land van Steijn 15 gulden), dochter van Herman (Harmen) van Grieken, veehouder en Jefje (Eva) Klare.

    Hij huwde 2e Oudewater 4 april 1774 (Jacob betaalt 15 gulden aan de gaarder in het Land van Steijn) met Aaltje Smit, geb. Oudewater. [Aaltje Smit huwde 2e Oudewater 31 december 1775 met Leendert Nielse, j.m. geb. Rotterdam].

    Neeltje van Grieken werd door haar moeder Eva Klare de boerderij Welgelegen (twee hofsteden met 29 morgen land) aan de Hogendijk in Stein geschonken. Haar zuster werd, zoals hierboven aangegeven, de boerderij aan de Goejanverwellesluis geschonken.

    Jacob Leeuwenhoek, wedr. van Neeltje van Grieken, verschijnt op 21 januari 1773 voor de weeskamer van Stein om zijn boedel te scheiden ten behoeve van zijn zoon Lambertus, voor wie als voogden optreden Gerrit en Adriaan Leeuwenhoek. Er is een inventaris van de boedel gemaakt. Jacob blijft in het bezit van alle goederen. Hij zal Lambertus opvoeden etc. en bij diens meerderjarigheid een bedrag van 4000 gulden uitkeren als moederlijk erfdeel. Als zekerheid gelden de twee hofsteden met 29 morgenland. Kort na het huwelijk met Aaltje Smit overlijdt ook Jacob Leeuwenhoek. De goederen van de boerderij Welgelegen worden op 13 maart 1775 op een boeldag verkocht, hetgeen 4033 gulden en 19 stuivers oplevert.
